There's evil lurking within the estate, as Verussa said, and they put the Bloodstone on it for the hunters to slay the monster and get the coveted title of being the leader. He's Man-Thing, who is he in Werewolf by Night and what are his powers? Also, how is he and Jack friends?
Who is Man-Thing in Werewolf by Night?
The hunters have prepared themselves for something sinister as part of the ritual of choosing the replacement of Ulysses Bloodstone is winning the coveted Bloodstone attached to a monster that they have to slay while every hunter looks after themselves so as not to get killed by the others.
The monster is also known as Man-Thing, a swamp-like creature with mushrooms and some kind of tentacles on its face. It is hideous and scary as the monster can incarcerate a human through its grip, leaving only the some remains of the person killed.
That being said, the Man-Thing isn't always totally evil. Yes, he may be a monster, given his monstrous appearance, there is still some good in him, a lot of it, actually. As it turns out, he's a friend of Jack Russell, and Jack seems to know the monster as "Ted."
Why is Man-Thing Called By Jack as Ted in Werewolf by Night?
Jack Russell seems to have uncovered a sweet spot for a monster like Man-Thing as he knows how to call him by the name "Ted." As it turns out, the only reason Jack joined the hunt is because he wants to free Ted and he doesn't even care about the Bloodstone.
In the comics, Man-Thing is actually Dr. Theodore Sallis, Ted, for short, and he was a biochemist who worked for one replicating the Super Soldier serum, yes, the one on Captain America, in a research called Project Gladiator.
A terrorist organization, Advanced Idea Mechanics or AIM, found out about the project and attacked the lab where Ted was working. To save his research and hopefully, himself, Ted injected himself the serum so no one could get ahold of it and then, his car crashed into a swamp.
Unfortunately for Ted, the swamp he fell into has a linkage to the Nexus of All Realities, an interdimensional gateway, and it affected the sample within Ted that resulted to his transformation into Man-Thing, a swamp-like creature with red eyes.

What Are The Powers of Man-Thing in Werewolf by Night?
As seen in Werewolf by Night, Man-Thing can incarcerate anyone with its touch should it will that such person needs to be eliminated, like what happened to Joshua Jovan, and Verussa, and this traces back to his comic book origins.
See, in the comics, Man-Thing could literally smell fear as he could sense emotions that when anyone is in fear and Man-Thing gets ahold of them, they would be burned. The series even has the tagline, "Whatever knows fear burns at the Man-Thing's touch!"
Man-Thing's abilities are a mix of the serum of Ted and the energies coming from the Nexus of All Realities. He is Immortal, strong, durable, invulnerable, and immune to any disease.
Despite all these, Man-Thing is a sweet being, taking care of Jack after transforming into a werewolf. Man-Thing even hands Jack a cup of coffee in the morning. Their friendship runs deep, as for one monster to another, they know what it is like to be misunderstood.
