A whole bunch of new faces just made their first appearances in The Last of Us Episode 4 and that includes Kathleen, leader of a movement in Kansas City. Even with the problem with the infected, she is focused on Henry and Sam but why is she looking for them?
Why is Kathleen Looking For Henry and Sam in The Last of Us?
The Last of Us Episode 4, titled Please Hold My Hand, just introduced new characters that would be pivotal for the narrative of Joel and Ellie moving forward with their journey to finding Tommy and bringing Ellie to a Firefly safehouse.
These new characters include Kathleen, who is first seen interrogating an old man who turns out to be a doctor and she throws him a ton of names of who may have ratted out her brother to FEDRA.
From this scene alone, Kathleen mentions Henry and she has been very persistent in finding him and his brother, Sam, for something yet to be revealed.

In the conversation between Kathleen and the doctor, there's already a hint as to why she is pursuing the people she mentioned, including Henry -- it is because of her brother and given that he isn't there, it's safe to assume that he died.
The people Kathleen mentioned may have ratted out her brother to FEDRA and he may possibly have fallen victim to the oppressive government. Kathleen may have won the war and took over Kansas City but nothing can fill the void of the loss of her brother.
With Henry being one of the people she is looking for, it could be deduced that Henry may have been involved with the death of Kathleen's brother, or could have been the reason of his demise. Kathleen's search is centered on one thing -- revenge.
A brother for a brother, possibly, Henry has Sam, his deaf younger brother with him and that could be Henry's biggest weakness that Kathleen could possibly exploit. She, herself, knows Henry wouldn't even let Sam starve.
