Arcane is set in a utopian region of Piltover and the oppressed underground of Zaun, and it follows the origins of two League champions and the power that will tear them apart. The two cities are located in the world of League of Legends, set in a continent called Runeterra. In the game, two teams of five players, who each control a particular Legend, work together to destroy the enemy's main structure known as Nexus.

Launched on PC in 2009, the free-to-play League of Legends is one of the most popular multiplayer video games in the world, and is a major e-sports title that amassed huge success for Riot Games, which is now one of the largest video game studios in the industry. The game's success had led to various spinoffs, including Legends of Runeterra, collectible card game, Teamfight Tactics, on iOS and Android, and League of Legends: Wild Rift, a mobile spin-off.
